In-locus C-term tagging
Native promoter retained. Cassette spans the junction. Limited to tags ≤ 36 nt: FLAG, His6, His8, Strep-II. For larger tags (3×FLAG, HiBiT, HA, c-Myc, V5, T7) use Workflow 03.

Plasmid expression
